Ocorre aproximadamente dez anos antes dos eventos da série original Star Trek, ST: Discovery mostra as casas unidas dos Klingons envolvendo-se em uma guerra com a Federação Unida de Planetas. Os protagonistas são os tripulantes da USS Discovery.

Na série, são contadas as aventuras da tripulação da nave USS Enterprise, que, no século XXIII, representa a Federação Unida de Planetas numa missão de cinco anos no espaço. O objetivo é explorar novos mundos, procurar novas formas de vida e civilizações. No fundo, chegar até onde o Homem ainda não havia chegado. Cada episódio da série começava com a frase: "Espaço, a última fronteira". A nave é comandada pelo Capitão James T. Kirk, interpretado por William Shatner, que tem como primeiro-oficial e melhor amigo Mr. Spock (Leonard Nimoy), um ser oriundo do planeta Vulcan. No grupo de personagens principais entram ainda o médico McCoy (De Forest Kelly). A tripulação da Enterprise ronda os 430 tripulantes. Ao longo da sua missão a Enterprise encontra e combate povos extra-terrestres e descobre uma série de novos mundos.

Dallas McKennon portrayed innkeeper Cincinnatus. Country Western singer-actor Jimmy Dean was a featured actor as Josh Clements during the 1968–1970 seasons. Actor and former NFL football player Rosey Grier made regular appearances as Gabe Cooper in the 1969 to 1970 season. The show was broadcast "in living color" beginning in fall 1965, the second season, and was shot entirely in California and Kanab, Utah.
